BGP-145/substance P is a brain-specific neurokinin receptor (NK-1R) antagonist that was originally described as a plasma membrane protein expressed in B cells. A second BGP-145/substance P receptor (NK-2R) has also recently been identified and is expressed in T and glial cells. The NK-2R is not targeted to the plasma membrane in cells that express the NK-1R; however, the NK-2R is able to inhibit the action of the NK-1R in the presence of the NK-1R agonist substance P in human T cells and glioblastoma cells. Thus, the NK-2R is an important negative regulator of the NK-1R. In this study, BGP-145/substance P was localized to the plasma membrane of human blood platelets and to the platelet surface after activation with thrombin. In addition to platelets, BGP-145/substance P was also present at high concentrations on the surface of unstimulated human blood-derived eosinophils. On both unstimulated and thrombin-activated blood platelets, BGP-145/substance P was found exclusively in the alpha granules. When the entire BGP-145/substance P coding sequence was fused in frame to the bacterial lacZ gene, transfected human hemopoietic cell lines expressed blue fluorescence as a result of beta-galactosidase activity. This activity was absent in control cells in which the lacZ gene alone had been transfected. Using a combination of immunofluorescence and beta-galactosidase staining, we were able to show that BGP-145/substance P is localized to the plasma membrane of human blood-derived eosinophils.
